现主要围绕动物病毒病,开展病毒与细胞互作、病毒感染与免疫的分子机制等基础研究,以及新型疫苗、抗体、病毒病快速诊断等应用技术研究,力争解析动物病毒感染与致病机制,建立有效疫病防控策略。新冠疫情期间,参与完成了新冠病毒溯源、抗病毒靶点筛选及疫苗研发。
2018-2025年,主持国家自然基金面上项目、国家转基因生物新品种培育重大专项(骨干)、国家重点研发(骨干)、吉林省科技发展计划(杰出青年、重点科技攻关)、长春市科技局重点研发、南平市科技特派员创新创业一等项目等10项课题,累计到位经费460余万元。授权专利3项。在PNAS、Front Immunol、Int J Biol Macromol等发表责任作者SCI文章50余篇(13篇中科院一区Top,2篇高被引文章,31篇中科院二区)。

(1)建立了病毒中和抗体基因筛选鉴定平台、基于腺病毒的外源基因高效表达系统、假型病毒制备平台以及可表达新冠病毒主要结构蛋白的假型病毒。
(2)建立了基于乳杆菌的外源基因表达系统,并分别制备了表达新冠病毒刺突蛋白及其受体结合区RBD基因的重组植物乳杆菌与病毒样颗粒(VLP),两种系统均可高效表达相应蛋白,可用于制备新冠病毒候选疫苗。
(3)建立了基因重组技术平台,可用于基因过表达或敲除研究;特别是建立了CRISPR-Cas技术体系,并用此技术制备了PRV感染抑制细胞系。
(4)建立了病毒标记技术平台,具有制备标记病毒的实验条件和能力。
(5)建立了免疫共沉淀、激光共聚集和SPR技术等蛋白相互作用技术平台,验证了PCV2主要蛋白Cap和Rep与细胞HMGCR互作的机制;阐明了TRIM21参与PCV2感染的机制。
(6)制备的IFITM3敲除小鼠模型,为解析宿主抗病毒分子机制提供平台。
(7)参与了新冠病毒溯源工作,分别总结新冠病毒检测方法、抗病毒药物及靶点、疫苗研究、宿主IFITMs抗病毒研究、外泌体研究的最新进展,并发表相关英文综述,及时地为新冠疫情的防控提供了理论依据及可行性方案。
(8)在PNAS发表的两篇文章分别从冠状病毒跨种传播、新冠病毒冷链传播两个方面进行了评述。提示我们应该重视并重新评估家畜和家禽病毒(特别是冠状病毒)的跨种传播风险,同时也应该重视物品(特别是冷链)传播新冠病毒的可能性,并采取必要的防范措施。
(9)完成SARS-CoV-2 S蛋白及其突变体的构建、突变体的表达与鉴定、突变体的假型病毒的包装与验证,并利用生物信息学对SARS-CoV-2 S蛋白突变体的蛋白稳定性进行了预测。
(10)围绕动物病毒病,致力于病毒与细胞相互作用、病毒感染与免疫的分子机制等基础研究。部分解析了猪圆环病毒2型(PCV2)持续性感染及免疫抑制的机理,为疾病防控提供了理论依据。
(11)发现PCV2能够感染人类细胞,且在感染后也能产生具有感染性的病毒颗粒;但是,PCV2对人源细胞的感染具有自限性,且该病毒对人源细胞的感染性较猪肾细胞PK-15低。为阐明PCV2是否会成为异种移植中潜在的人兽共患病毒提供理论依据。
(12)首次发现水貂圆环病毒也可感染狐狸与貉子。揭示动物圆环病毒具有宿主广、共感染现象明显等特点。
(13)国内外首次成功拯救出PCV4,并研制出PCV2/PCV3/PCV4治疗性抗体及相关三价疫苗。
